Protein medications are the most rapidly expanding class of therapeutics, serving patients with diabetes, cancer, cardiovascular, renal, gastrointestinal, rheumatologic and neurological diseases, among many others. The therapeutic and commercial value of pioneering drugs including insulin, erythropoietin, G-CSF, plasminogen activator, and interferons is undisputed, yet second generation medications have improved the therapeutic efficacy of these parent products, by enhancing their performance in potency, time action, and other properties.
As injectable medications, natural versions of bio-therapeutic proteins are simply not designed for best performance. The physical, chemical and pharmacological properties are sub-optimal. Current protein engineering approaches have been limited to shuffling the position and identity of 20 amino acids.
